The PathHunter U2OS MC4R(T162I) Pharmacotrafficking Assay is a stable, engineered cell line for use in studying pharmacochaperones targeting MC4R. This cell line enables assessment of chaperone-based stabilization of MC4R(T162I) via detection of rescued membrane-endosome trafficking. The included cell line overexpresses ProLink-tagged mutant MC4R(T162I), which is misfolded and retained in the ER. The cell line also expresses endosome-localized EA. When a pharmacochaperone stabilizes the mutant MC4R, it is trafficked to the membrane and then internalized to the endosome, producing EFC signal.
